THE EFFECT OF KETAMINE ON TRANSMEMBRANE POTENTIALS OF PURKINJfi FIBRES OF THE PIG HEART

چکیده

The action of ketamine upon spontaneously active and electrically driven Purkinje fibres of the moderator band of the pig heart has been studied using a floating microelectrode technique. Ketamine hydrochloride altered the transmembrane potential in a dose-related and reversible manner. Concentrations of 1X 10M were subthreshold whereas 5 X 10'M and 1X 10~M slowed the frequency and increased the action potential duration of spontaneous preparations, actions consistent with an antiarrhythmic effect. Higher concentrations of 5 X 10'M ketamine initially led to a shortening of the action potential duration, then failure of the original spontaneous activity. During recovery pacemaker-like activity developed associated with a loss of resting membrane potential. In electrically driven preparations 5 X 10"M ketamine significantly shortened the duration of the action potentials between evoked potentials and markedly augmented the response to adrenaline, actions consistent with an arrhythmogenic effect. These findings suggest a basis for isolated reports of cardiac side effects.

Effect of Aqueous-Ethanolic Extract from Rosa damascena on Guinea Pig Isolated Heart

Objective(s) In the present study, the effects of aqueous-ethanolic extract from Rosa damascena on heart rate and contractility were examined. Materials and Methods Isolated guinea-pig hearts were perfused through aorta in a Langendorff mode. Heart rate (HR) and contractility were determined in the presence of four concentrations of the extract (0.1, 0.2, 0.4 and 1.0 mg %) and isoprenaline (...

The Role of Nitric Oxide and Prostaglandins in the Effect of Adenosine on Contractility, Heart Rate and Coronary Blood Flow in Guinea Pig Isolated Heart

It is a well-established fact that adenosine and its receptor subtypes (A 1 and A ) are involved in changes of contractility, heart rate and coronary blood flow (CBF) under different circumstances. This study was conducted to evaluate the role of nitric oxide and prostaglandins in development of these changes. For this purpose, Nitro-L-Arginine methyl ester (L-NAME), and indomethacin as inhibit...

The effect of ketamine on synaptic transmission and synaptic plasticity in CA1 area of rat hippocampal slices

The effect of ketamine (1-100 µM), which has NMDA receptor antagonist properties, on synaptic transmission and long-term potentiation (LTP) in CAl area of rat hippocampus was examined in vitro. Field potentials were recorded in pyramidal cell layer following Schaffer collateral stimulation. Primed-burst stimulation (PEs) was used for induction of LTP. The amplitude of population spiks (PS) was ...
